GENERAL INFLAMMATION MODELS
Induction of general inflammation in mouse or rat models is useful to evaluate the
anti-inflammatory effect of a test article. As a read-out of the therapeutic effcacy of the test article, samples can be isolated for histology, cytokine levels or gene expression analysis. Please refer to our In-Vitro services section for more information on ex-vivo studies oered by SIA.
LPS Model
DTH MODEL
Delayed-type hypersensitivity (DTH) model can be used for rapid screening of anti-inflammatory compounds. The DTH-model is performed by sensitizing mice with a sensitizing agent (e.g. Oxazolone) on the abdomen and a week later challenge of the ears with the sensitizing agent. An acute DTH response occurs on the ears and can be followed by measuring the ear thickness, which is signicantly increased and
sustained for up to 7 days.
LPS MODEL
Sepsis is a systemic inflammatory condition associated with the presence of pathogenic microorganisms or their toxins in the blood. Symptoms associated with the presence of circulating endotoxin include fever, hypertension, and, under more extreme cases, endotoxic shock and death. Lipopolysaccharide (LPS) is an endotoxin, an outer membrane component of Gram negative bacteria involved in the sepsis pathology that is responsible for toxic activity.
LPS challenge leads to acute inflammation and elevation of the pro-inflammatory cytokine levels.
